Grain spawn: how to grow it, step by step

Between a clean culture and a full substrate sits a jar of colonised grain. Rinsing, the 12-hour soak, sterilising at 121 °C, and the 10–14 days after.

Grain spawn is the least glamorous and most decisive step in growing mushrooms. It is grain, usually wheat, rye, or corn, that a mushroom mycelium has grown all the way through, turning each seed into a nutrient-loaded starter you can scatter into a larger substrate. Get it clean and evenly colonised and the rest of the grow tends to follow. Rush the boil or cut a corner on sterility and you find out days later, when green mould answers instead of white mycelium.

Where it sits in the chain

Cultivation runs from a small, clean culture to a large fruiting substrate, and grain spawn is the link in the middle. You start with a clean culture on an agar plate or in liquid, take a tiny piece of it, and grow that piece across a jar of sterilised grain. The colonised grain then goes on to inoculate the bulk substrate. The grain does two jobs at once. It multiplies a pinhead of clean mycelium into hundreds of separate growth points, and it carries nutrients into the substrate so the fungus starts fast instead of starving while it settles in.

Mycelium grows only at the tips of its threads, the hyphae, which run about 1.5 to 10 micrometres thick. That is why more starting points matter so much. Colonisation spreads outward from every grain at once, so a well-mixed jar closes over far quicker than a single lump of culture would.

Preparing the grain

Use quality grain and rinse it several times in cold water until the water runs clear. You are washing off dust, chaff, and loose starch, all of which feed contaminants later. Then soak the grain at roughly one part grain to two parts water for about 12 hours so it swells and takes up moisture evenly.

Rye and millet are the usual favourites. Their small, hard kernels stay separate and give many points of contact, but wheat, barley, corn, and sorghum all work too. The choice matters less than the moisture: the finished grain should be plump and hydrated, never sitting in a puddle.

The boil

Simmer the soaked grain for 30 to 40 minutes until it is soft but the kernels are still whole. If they split and spill their starch, they turn gummy and clump, and clumped grain colonises unevenly. Drain thoroughly, then spread the grain out and let it cool and surface-dry. This last part is easy to skip and it matters. Waterlogged grain suffocates the mycelium and is a standing invitation for bacteria.

Sterilising the jars

Fill glass jars of 0.5 to 1 litre about two thirds full with the cooled grain. Leave the top third empty so you can shake the jar later without packing everything solid. Close each jar with a filtered lid: a hole plugged with cotton or synthetic wool lets the mycelium breathe while keeping spores out.

Sterilise in a pressure cooker or autoclave for 1.5 to 2 hours. The standard is about 15 psi, which corresponds to 121 °C. This is the step that clears the field. It kills the bacteria and mould spores riding on the grain so the mushroom you introduce has no competition. Grain spawn is sterilised, not merely pasteurised, because it is so nutritious that any surviving microbe would race the mushroom for the same food and usually win.

No autoclave? Use a large pot, but repeat the process on two or three consecutive days. This is fractional sterilisation, or tyndallisation. Each round kills active cells but not all heat-resistant spores. Between rounds, at room temperature, those spores germinate into vulnerable cells that the next boil finishes off. It is slower and less certain than a pressure cooker, and it is the honest fallback when you do not have one.

Inoculation

Let the jars cool to room temperature before you open them. Warm grain has a rising column of air above it that pulls in room spores, and heat can kill the culture you are about to add. Work in still, sterile conditions, next to a flame or inside a still-air box, and introduce the mycelium from your agar wedge or liquid culture. Then shake the jar so the inoculum spreads through the grain rather than sitting in one spot.

This is the moment contamination gets in, so slow down here. Clean hands, flamed tools, a wiped surface, minimal air movement. Everything you did in the boil and the steriliser can be undone in the ten seconds a lid is off in a draughty room.

Incubation

Keep the jars at 20 to 25 °C, out of direct light. Over roughly 10 to 14 days the white mycelium runs from every grain and knits the jar into one solid mass. A day or two in, give it a gentle shake to redistribute the growth and speed things up. When the grain is fully white, with no untouched kernels and no off-colours or smells, it is ready.

Watch the jar rather than the calendar. Colonising bulk substrate afterwards usually takes another two to three weeks at a similar temperature, and species differ. Indoor grain spawn closes in days to weeks, while an outdoor log inoculated with the same fungus can sit for a year or more before it fruits. The numbers here are for indoor grain, and they are a guide, not a stopwatch.

Storing finished spawn

Fully colonised spawn keeps in the fridge for one to two months, no more, and around +2 °C holds it best. Cold slows the mycelium without killing it, but colonised grain slowly loses vigour, and if any contaminant slipped in, time is exactly what it needs to show itself. Use spawn while it is fresh and white. If you see a spot of another colour or catch a sour smell, do not gamble on it: throw the jar out and start clean.

The bridge, and why patience wins

Grain spawn earns its place by being the bridge. It takes a fragile scrap of clean culture and turns it into enough vigorous, evenly spread mycelium to conquer a whole substrate. Nothing about it is magic. It is clean grain, real heat, a still moment at the lid, and the patience to let the mycelium grow at its own pace. That plain, unhurried work is most of the craft, and it is the same honest process behind the finished extracts we make.
